Uses

  • Nerve pain – diabetic neuropathy, post-herpetic (shingles) pain, sciatica-type pain.
  • Preventing migraine and tension-type headaches.
  • Depression, usually at higher doses.
  • Poor sleep linked with chronic pain or fibromyalgia.
  • Bed-wetting in children, on a paediatrician's advice.

How to take it

Take one 10 mg tablet once a day in the evening, 1 to 2 hours before bedtime, with or without food. Swallow it whole with water. For pain and migraine, your doctor usually starts at 10 mg and may increase slowly every week or two; the benefit for pain takes 2 to 6 weeks to appear. Expect drowsiness and a dry mouth at first – avoid driving until you know how it affects you. Do not stop suddenly after long use; the dose is reduced gradually.

Safety information

Alcohol

Avoid alcohol. It greatly increases drowsiness and the risk of falls, and an overdose with alcohol is dangerous.

Pregnancy

Tell your doctor if you are pregnant or planning. It is used in pregnancy only when needed. Small amounts pass into breast milk; discuss with your doctor.

Kidney problems

No routine dose change, but tell your doctor about kidney disease.

Liver problems

Cleared by the liver; lower doses are used in liver disease.

Children

Not used for depression in children. Used for bed-wetting or pain only under a paediatric specialist.

Missed dose

If you remember at bedtime, take it. If it is already morning, skip it – a late dose will make you drowsy all day. Never double the dose.

Interactions

  • Other antidepressants (fluoxetine, sertraline, escitalopram), tramadol – serotonin syndrome and higher amitriptyline levels.
  • MAO inhibitors – dangerous; a gap of 2 weeks is needed.
  • Sleeping tablets, alprazolam, codeine, sedating antihistamines (cetirizine, chlorpheniramine) – much more drowsiness.
  • Blood pressure medicines like clonidine – effect may be blocked; amlodipine and others – more dizziness.
  • Heart rhythm medicines, antipsychotics, some antibiotics (azithromycin, levofloxacin) – heart rhythm effects.
  • Antispasmodics, dicyclomine – more dry mouth, constipation and urine retention.

Frequently asked questions

When should I take Tryptomer 10?

In the evening, 1 to 2 hours before bed. It causes drowsiness, which helps sleep, and taking it earlier reduces the morning hangover feeling.

Is Tryptomer 10 a sleeping pill?

No, though doctors use its sedating effect to help sleep in people with chronic pain. At 10 mg it is mainly prescribed for nerve pain, migraine prevention and sleep, not as a routine sleeping tablet.

Can I take Tryptomer 10 with alcohol?

Avoid it. Alcohol multiplies the drowsiness and dizziness and raises the risk of falls and accidents.

How long does Tryptomer take to work for nerve pain?

Sleep may improve within days, but pain relief usually takes 2 to 6 weeks, and your doctor may increase the dose gradually. Do not stop early because it has not worked yet.

Is Tryptomer 10 addictive?

No. It does not cause craving. Stopping suddenly after long use can cause withdrawal symptoms like nausea and sleeplessness, so the dose is reduced slowly.

Is Amitone 10 safe in pregnancy?

Tell your doctor if you are pregnant or planning. It is used only when the benefit clearly outweighs the risk, and other options are often tried first.

Can I take Tryptomer 10 with painkillers?

Paracetamol and most NSAIDs are fine. Avoid tramadol without advice, as it can cause serotonin syndrome and fits with amitriptyline.

Tryptomer 10 vs Tryptomer 25 – what is the difference?

Only the strength. 10 mg is the usual starting dose for pain and migraine; 25 mg and higher are used if needed or for depression. Your doctor increases the dose step by step.

Why is my mouth so dry on Tryptomer?

Dry mouth is one of the commonest effects. Sip water, use sugar-free gum and keep up dental care. It often eases after a few weeks; tell your doctor if it does not.
